Soybean (Glycine max [L.] Merr.) cell suspension cultures (cv. Williams 82) inoculated with the pathogenic bacteria Pseudomonas syringae pv. glycinea respond with a hypersensitive reaction (HR) when the bacteria express the avirulence gene avrA. A mRNA differential display was established for this system to allow the identification of genes induced during the HR. Six PCR-fragments (DD1–DD6) from the differential display analysis were identified, which are induced during the HR. Database searches revealed that the fragment DD1 encodes chalcone isomerase and DD2 was identified as ubiquitin. The fragment DD3 shares significant homology to the signalling molecule 14-3-3. The partial DD4 product is homologous to the enhancer of rudimentary from Drosophila and an uncharacterized homologue of it from Arabidopsis. The fragment DD5 is similar to glucose-6-phosphate dehydrogenase which provides NADPH to the cell. The PCR-product DD6 seems to be a new leucine-rich-repeat disease resistance gene from soybean, which is significantly induced during the HR. All of the identified genes are clearly induced during a HR in infected plants of the same cultivar, indicating that results from the cell culture model system can be transferred to intact plants. These studies show that complex mRNA differential display is a powerful tool to identify new induced gene in plant-pathogen interactions. |

Resumo: O objetivo deste estudo foi avaliar as variáveis hematológicas de tilápias para obtenção de potenciais biomarcadores de peixes no monitoramento de qualidade da água dos sistemas de aquários experimentais. Avaliou-se as alterações na série vermelha em juvenis de tilápias mantidas durante 30 dias em sistema de aquários com volume útil de 300 L provenientes dos grupos controle de dois experimentos conduzidos no Laboratório de Ecossistemas Aquáticos da Embrapa Meio Ambiente. No experimento 1 utilizaram-se quatro tratamentos e três repetições em sistema sem recirculação. No experimento 2 utilizaram-se quatro tratamentos e cinco repetições em sistema com recirculação da água. Os parâmetros de monitoramento da água dos dois experimentos, como o pH, o oxigênio dissolvido, a temperatura e condutividade foram mensurados diariamente e apresentaram valores sem diferença significativa entre os dois experimentos. No entanto, o teor de amônia total foi mais alto no sistema sem recirculação da água. Desta forma, observou-se que os peixes mantidos no sistema com recirculação de água apresentam respostas hematológicas de acordo com o descrito na literatura e os peixes do sistema sem recirculação apresentam valores mais baixos de hemoglobina, hematócrito e eritrócitos. Alterações na série vermelha em juvenis de tilápias observadas nos experimentos avaliados neste trabalho indicam seu potencial como biomarcadores de qualidade da água. Abstract: The aim of this study was to evaluate the hematological variables of tilapia to obtain potential biomarkers of fish in the monitoring of water quality of experimental aquarium systems. The changes in the red series in juveniles of tilapia kept for 30 days in a system of aquariums with a useful volume of 300 L from the control groups of two experiments conducted at the Laboratory of Aquatic Ecosystems of Embrapa Environment were evaluated. In experiment 1, four treatments and three replications were used in a system without recirculation. In experiment 2, four treatments and five replications were used in a system with water recirculation. The water monitoring parameters of the two experiments, such as pH, dissolved oxygen, temperature and conductivity were measured daily and presented values with no significant difference between the two experiments. However, total ammonia content was higher in the system without water recirculation. Thus, it was observed that the fish kept in the system with water recirculation present hematological responses as described in the literature and the fish of the system without recirculation present lower values of hemoglobin, hematocrit and erythrocytes. Changes in the red series in tilapias observed in the experiments evaluated in this work indicate their potential as biomarkers of water quality.
